ATF6-GFP reporter nuclear translocation in VACV infected MEFs. A31 MEFs were transfected with the pCMVshort-EGFP-ATF6α and infected with VACV strains for 24 h. Cells were carefully collected and lysed with nuclear isolation buffer. Isolated nuclei obtained after washing were analyzed for green fluorescence in a FACSCan flow cytometer. 10.000 events were collected inside the gate determined for nuclei size and granulosity. Results were plotted as histograms for green fluorescence intensity versus cell counts
