Optimising RT-LAMP temperature. RT-LAMP reactions for ORF 8 (a) and N (b) detection of SARS-CoV-2 were incubated at 63 °C, 65 °C and 67 °C for 35 min. Extracted RNA from SARS-CoV-2 infected Vero cell culture supernatant was tested along with 1/2, 1/4 and 1/16 dilutions in PCR grade water. As negative controls, pooled negatively tested patient RNA (Negative) and water (NTC) were used. All experiments were performed in triplicates three times over.
