Cells were mock treated, and TNF-α (10ng/mL) stimulated, or infected with either Thiverval or GXW-07 at an MOI of 1 at 48 h p.i. in PK-15 cells. Nuclear extracts were prepared and subjected to EMSA with a Digoxin labeled NF-κB probe. Anti-p65 rabbit polyclonal antibody was used in supershift assay. A 100-fold molar excess of unlabeled NF-κB probe (unlabeled competitor) was added to analyze the specific bands.
