Two M protein peptides stimulate IFNγ+ production in splenocytes from immunized BALB/c mice in an ELISPOT assay. Splenocytes from immunized BALB/c mice were (A) left unstimulated, (B) stimulated with an irrelevant peptide, (C) stimulated with the peptide "F57G YMTFVHF": 54 spots/5 × 105 cells, (D) stimulated with the peptide "K93F ITSRCRL": 81 spots/5 × 105 cells, or (E) stimulated with PMA and ionomycin (324 spots/5 × 105 cells) to determine the frequency of IFNγ-secreting splenocytes.
