Principle and evaluation of the RVFV-Ren antiviral assay. (A) Schematic outline of the RVFV-Ren antiviral assay. (B) Dose-response curve in human A549 cells. A549 cells seeded in 96-well plates were treated with increasing doses of Multiferon for 7 hours before infection with 10,000 plaque forming units of RVFV-Ren. Sixteen hours later, cells were lysed and Renilla luciferase activity in cell lysates was determined. All measurements were performed in triplicate wells under standard conditions; shown are means ± SD. Both axes are plotted in logarithmic scale; the dotted vertical line indicates the threshold of sensitivity, based on the mean + 2SD of an untreated control run in triplicate; shaded area shows the linear range of the assay; RLU, relative light units.
