Use of rNTPs by wild-type RTs and their chimeric derivatives. The ability of reverse transcriptases from the wild type HIV-1 and MuLV and their chimeric derivatives to incorporate rNTPs was examined on 49-mer U5-PBS DNA (Panel A) and U5-PBS-RNA (Panel B) templates primed with the 5'-32P-labeled 17-mer PBS DNA primer. Reactions were done at 37°C for 30 min as described in Materials and Methods. Lanes 1 and 2 in each panel represent extension reactions done in the presence of 500 μM of dNTPs and rNTPs, respectively.
