Interferon action on the IRES-mediated translational inhibition was prevented in cured resistant cell lines. Effect of interferon on IRES-GFP translation regulation was examined using one cured sensitive and nine resistant Huh-7 cell lines. Cells were transfected with IRES-GFP plasmid and treated with 1000 IU/ml interferon alpha. After 24 hours, GFP expression was recorded. Cured cells prepared from resistant clones unable to activate interferon signaling and no inhibition of HCV IRES was seen.
