PTD tat modification of Ad5 fiber enhanced in vivo gene delivery efficacy of the vector. In vivo gene delivery of Ad5.PTDtat was examined using a non-invasive fluorescence imaging technique in low-CAR expressing tumor models. 1010 VPs of Ad5 or Ad5.PTDtat were injected into the subcutaneous U118MG tumors, and in vivo green fluorescence images were acquired at different days post viral injection. (A) Representative in vivo images from PBS, Ad5, or Ad5.PTDtat injected mouse tumor models at day 7 after vector administration. The colors representing different intensities of signal are shown on the color bar. Ad5.PTDtat infection resulted in more intensive GFP signals than unmodified Ad5 vectors.
