ZNF148 suppresses the expression of RXRα. (A) The mRNA levels of various relevant transcription factors in ZNF148-overexpressing Huh7 cells were examined by real-time PCR. (B) The mRNA level of RXRα in HepG2-NTCP and Huh7 cells infected with lentivirus expressing shCont/shZNF148 was measured by real-time PCR. (C-D) The protein level of RXRα in HepG2-NTCP and Huh7 cells transfected with the vector/ZNF148 plasmid or infected with lentivirus expressing shCont/shZNF148 were determined by Western blotting. (E) Huh7 cells were transfected with the vector/Flag-ZNF148 plasmid. After 72 h, the cells were harvested and subjected to immunoprecipitation with IgG or an anti-Flag antibody. The level of the RXRα promoter was measured using real-time PCR. (F-G) A dual luciferase reporter assay was performed to test the activity of the RXRα promoter. *P < 0.05, **P < 0.01
