Effect of monovalent and divalent cations in the alteration of HAV by soluble HAVCR1. Sedimentation analysis of cell culture-adapted HAV treated with soluble HAVCR1 in the presence or absence of different cations. Purified HAV virions were incubated with 5 μg of D1 muc-Fc for 2 h at 37°C in the presence of 1 mM CaCl2, 10 mM MgCl2, 0.1 mM ZnCl2, 0.1 μM MnCl2, 500 mM NaCl, or 10 mM LiCl and also in the absence of any cations, and sedimented in linear 15 to 30% sucrose gradients as described in Figure 2. HAV antigen was detected by ELISA. The data are mean OD 450 nm from duplicate wells; duplicate values varied by less than 10%. Poliovirus native virions and empty particles labeled with 35S-methionine were used as 160S and 80S sedimentation markers.
