The depletion of Cohesin components affects HSV-1 production. a, b Human primary fibroblast BJ cells were infected with lentivirus containing control shRNA or shRNA targeting SMC1 or Rad21 for 48 h (a). HeLa cells were transfected with control siRNA or siRNA targeting SMC1 or Rad21 for 48 h (b). Then cells were infected with HSV-1 at an MOI 5. Genome copy numbers were detected at 0, 2, 4, 6, 8, and 10 h post-infection (hpi). Shown is DNA fold-change over control at 0 hpi. Error bars represent the standard deviation from three independent experiments. c HeLa cells were first knock-down SMC1 or Rad21 with siRNA, then were infected with HSV-1 at an MOI 0.1 and harvested cells and virus at various times post-infection.
