Knockdown of HDAC1 expression in HIV-1 producer cells affects virion infectivity in a cell type-dependent manner. 293 T cells were transfected with 20 nM siHDAC1 (Hs_HDAC1_6) or control siRNA (GFP-22 siRNA). At 48 h post-transfection, cells were transfected with pNL4–3 or co-transfected with pNL4–3.Luc.R-E- and pCMV-VSV-G vectors to produce wild-type or pseudotyped virions, respectively. Culture supernatants were harvested at 48 h after proviral DNA transfection and measured for virus production by p24 ELISA. Equal amounts of pseudotyped NL4–3.Luc.R-E- virions (p24 levels) were used to infect HeLa-CD4-LTR-β-gal (30 ng p24 per 105 cells) (a), Jurkat (30 ng p24 per 106 cells) (c) or primary CD4+ T cells (30 ng p24 per 106 cells) (d). b LuSIV indicator cells were infected with NL4–3 virions (30 ng p24 per 106 cells). Infectivity was measured in terms of luciferase activity and determined at 48 h post-infection. Results were calculated as a percentage of the control and represent means ± SD of six measurements from a single experiment (n = 6). Results are representative of two to three independent experiments and were analysed by the Student t test (*, p < 0.05, **, p < 0.01)
