From: A preliminary analysis of hepatitis C virus in pancreatic islet cells

Total RNA from 3 different human islets donors was isolated using TRIzol reagent in combination with the RNeasy Mini kit followed by DNase treatment. 500 ng of total RNA were retrotranscribed using the Superscript III kit. The cDNAs obtained after retrotranscription were used as templates for quantitative real-time RT-PCR for mRNAs corresponding to the HCV entry factors CD81, occludin, claudin-1, and SR-B1. The relative amount of specific mRNA was normalized to Glyceraldehyde 3-phosphate dehydrogenase (GAPDH). mRNA levels in Huh7.5 cells and human islets are relative to those in 293 T cells.
