Perforin is produced by CD8+ T lymphocytes in the spleen of C57BL/6 mice after infection. Groups of C57BL/6 (WT) and TLR2/9−/− (KO) mice (6 animals/group) were infected with 106 p.f.u. HSV-1 via the intranasal route and, on the 5th day post infection, mice were euthanized, and TG and spleen cell suspensions prepared for flow cytometry analysis. The frequency of each cell within the live leukocyte population was determined. a Frequencies of perforin-producing CD4+ T cells (CD45+CD3+CD4+); b CD8+ T cells (CD45+CD3+CD8+); c NK T cells (CD45+CD3+NK1.1+) and d NK cells (CD45+NK1.1+).
