ATM and ATR siRNAs moderately reduce viral DNA production. HFFs were transfected with siRNAs targeting ATM or ATR. At 72 h post-transfection, cells were infected with HSV-1 or HSV-2 (MOI 5) and harvested at 16 h p.i. a ATM and ATR transcript levels in siRNA-transfected cells were quantified using qPCR, normalized to 18S and control siRNA (Allstar, AS) by the 2–ΔΔCt method. b Protein lysates of siRNA-transfected cells were immunoblotted and probed with antibodies specific for ATM or ATR. c Viral DNA levels (vDNA) were quantified by qPCR with ICP0 primers, normalized to 18S rDNA levels and Allstar control siRNA. d HSV-1 DNA levels in HFFs transfected with pooled ATM and/or ATR siRNAs were quantified by qPCR. In panels (a), (c), and (d), grey lines indicate the average of three experiments. Asterisks indicate statistical significance relative to Allstar control at p < 0.05
