Poliovirus infection inhibits poly(I:C) induced IRF-3 activation. a. Experimental design. At zero hours cells were either mock infected or infected with poliovirus at an MOI of 100 and at 1.5 h following infection or mock infection cells were either treated or not with poly(I:C). After another 1.5 h/ 2.5 h lysates were harvested to extract protein. b. HEC-1B cells were either mock-infected or infected with poliovirus at an MOI of 100 and then treated with poly(I:C) or not for the indicated amount of time. Whole cell lysates were prepared in non-denaturing lysis buffer and were analyzed by native PAGE followed by immunoblot to detect IRF-3. The monomeric and dimeric forms of IRF-3 are indicated. hpi: hours post infection
