Characterization of the Nsp2 domains responsible for inducing NF-κB activation.

(A) Schematic representation of various truncated mutants of PRRSV Nsp2 used in this study. PL2, the putative enzyme domain; HV, the hypervariable region; TM, the transmembrane region. (B) HeLa cells were co-transfected with the 0.2 μg of pNF-κB-Luc, 0.05 μg of pRL-TK, and the distinct truncations of Nsp2 expression plasmids (0.6 μg). An empty vector was used as a control. At 30 h after transfection, one group of cells with empty vector transfected was stimulated with 20 ng/mL of TNF-α. Cell extracts were collected at 36 h after the initial transfection and analysed for luciferase expression. **P < 0.01 as compared with vector control.
