The effect of HPV oncogenes on the transcriptional activity of different promoters in HFK cells. (A) HFK cells were co-transfected with 0.5 μg of the p53 reporter construct p53-Luc, along with 0.25 μg of either control vector (pcDNA) or 0.25 μg of expression constructs encoding HPV16 E6 or E7 or both expression vectors (0.125-0.125 μg) as indicated. (B) HFK cells were co-transfected with 0.5 μg of the adenovirus E2 reporter construct pAdE2Luc, along with 0.25 μg of either control vector (pcDNA) or 0.25 μg of expression constructs encoding HPV16 E6 or E7 or both as indicated. (C) HFK cells were co-transfected with 0.5 μg of the IVL reporter construct pGL3-IVL, along with 0.25 μg of either control vector (pcDNA) or 0.25 μg of expression constructs encoding HPV16 E6 or E7 or both as indicated. After transfection, HFK cells were either left in serum-free medium or induced to differentiate in DMEM (with serum and high calcium). The luciferase activities are shown relative to the activity of cells co-transfected with reporter constructs and the empty expression vector (pcDNA).
