Expression from capped transcripts containing both UTRs of segments 1, 4, 6, 8 or 10 is responsive to NS1. (A) The 5 + 3 UTR constructs contain the 5 UTR and 3 UTR of BTV-1 segments 1 (VP1), 4 (VP4), 6 (NS1), 8 (NS2), or 10 (NS3) immediately flanking the Renilla luciferase ORF. The 5 end of the capped reporter RNAs are defined by a T7 promoter and the 3 end is determined by a Bsm BI site, as described in Figure1. (B) BSR 96 well monolayers transfected with 100 ng pCAG NS1 and capped reporter transcripts. The amount of luciferase was assayed in triplicate 20 h after transfection with the reporter RNAs. Bars indicate one standard deviation from the mean. Fold of NS1-dependent upregulation indicated.
