Specific T cell responses were tested directly ex vivo in IFN-γ Elispot assays against peptides HPV16E7 49-57 and HPV18E6 67-75 . C57BL/6 mice (n = 4 per group) were immunised with PBS, rVVJ1175, rVVJ16E6E7, rVVJ18E7E6 or rVVJ16/18E7E6. Five hundred thousand splenocytes were incubated ex vivo with or without 5 μg/ml of HPV16E749-75 or HPV18E667-75 peptides. Plates were incubated for 24-28 hours at 37°C. Background IFN-γ production was generally found for the medium without peptides to be below 5 spots/million splenocytes. Asterisks represent statistically significant differences relative to the rVVJ1175 virus control (* p < 0.05, ** p < 0.001). Data shown are means ± standard deviations for three independent experiments.
