Construction of rLSDV-grttn. (a) transfer vector pYS-05. LSDV infection (0.1 ffu/cell) and pYS-05 transfection (100 ng DNA) of FBT cells and detection 24 hours later of (b) GUS using standard staining procedures and (c) Grttn in the cytosol of the cells by immunofluorescence using sheep anti-p24 and FITC conjugated anti-sheep. (d) Western blot detection of Grttn in cell lysates using sheep anti-RT and alkaline phosphatase conjugated anti-sheep.
