Expression of wild type avian NS1 reduces Interferon-induced STAT1 activation. Upper panel. Western blot analysis of 293 cells transfected with plasmids expressing wild type avian NS1 (A), wild type human NS1 (H), non-PDZ binding avian NS1 mutant (Aa) or vector alone (C). Cells were treated for 5 h with 1 × 104 U/ml Hplc-purified Interferon-α prior to harvesting. The blot was probed with anti phospho-STAT-1 antibodies to detect activated STAT1. Lower panel. The blot was reprobed with anti-α-actinin antibody to control for cellular protein input.
