Schematic diagram of the four DNA vaccine constructs. (A) pV-H3HA, (B) pV-H1HA1, (C) pV-H3-H1, (D) pV-H3-EHA-H1. The Kozak sequence was added before the ORF to promote protein expression. The MHC class II-restricted epitope box, EHA, was inserted into the co-expression plasmid pV-H3-H1. A flexible linker (G4S)3 was added to allow effective fusion gene expression and promote the correct folding of expressed proteins. The autocleaving 2A protein linker from foot and mouth disease virus was also added to allow cleavage of the fusion protein after expression.
