Ad5.PTD tat exhibited enhanced gene transfer efficacy in a variety of tumor cells. Gene transfer efficacy was evaluated by use of a GFP reporter that was carried in the E1 region of each vector. In the assay, tumor cells expressing varying levels of CAR were infected with either Ad5 or Ad5.PTDtat at an MOI of 100 or 500 VPs/cell, and GFP expression was examined by fluorescence microscopy and a fluorescence plate reader. (A) Representative fluorescence images of low-CAR containing cells (RD), medium-CAR containing cells (D65MG) and high-CAR expressing cells (Hela) that were infected with Ad5 or Ad5.PTDtat at an MOI of 500 VPs/cell. (B) GFP expression in a variety of cells infected with either Ad5 or Ad5.PTDtat was quantified using a fluorescence plate reader.
