Ad5.PTD tat showed similar CAR-binding activity to unmodified Ad5 vector in an ELISA-based binding assay. In the experiment, 109 VPs of each viral vector were immobilized in the wells of a 96-well ELISA plate, and incubated with increasing concentrations of recombinant sCAR (extracellular domain of CAR, i.e. soluble CAR). The binding activity was detected by AP activity conjugated to detection antibodies.
